You don't have to use pre-mix, but you should ensure the pump is accurately set. There is a possibility of fouling the plugs at lower engine speeds without the solenoid control, so ensure you only use the highest quality, low-ash/clean burning fully synthetic 2T oil, and that the jetting optimally set. (Please do not ask for/discuss jetting settings in the public forums. HRC base settings are available in the Tuning section of the main site.)
You won't be able expect to just jump on it and ride it like a standard road bike, however there is no reason not to still use the pump if you want to. We've set up plenty of dual purpose street/track bikes with oil pumps, and never had a problem. The downside on a track bike is the weight and possible failure of the pump (I've personally never seen an NSR pump fail in 30 years), and the effect 2T injection can have on overall fuelling. Pre-mix is far more controllable.
We've long recommended and used Rock Oil Synthesis 2 Injector*†. If this isn't available in your region, then look for a 2T that meets the same specifications.
*We use Rock Oil Synthesis 2 Racing in the NSR500Vs. "Racing" is not suitable for injection.
†We are not sponsored or financially involved with Rock Oil in any capacity. Other oils are available.
