I cant make up my mine about sprockets and chain pitch, 428 or 520 ?
The original is 520 pitch but seems quite a heavy chain and a bit over the top for a 250cc bike .
I understand the non o-ring on an MC21 as it has a cush drive... But on an MC28 which doesn't have a cush drive I'm wondering if having an o-ring chain helps with the harshness of the transmission?

I was of the impression the O'ring is only for lubrication and longevity of the chain and did nothing for cushioning drive train lash from acceleration/gear changes etc.

That's correct, as you will see the rings are on the side of the chain.

Totally agree, and the other downside for the non o-ring is that it won't last as long as the o/x-ring chains.
Soooo,
Road bike = o/x-ring chain
Race bike = Non o/x-ring chain
Who remembers Rossi crouching down next to his Aprilia 250 putting the chain back on at the side of the track. I believe that was a #428.
The Stop & Go team used both #520 and #428 in their Moto2 campaign last year. It wasn't so much a quest for more power to the wheel, but more about fine tuning the gearing.
